I got this rear brake new but it looks like there's a barb in the hose already but not sure since there's one in the box too. Cheers

Nope, no barb in that hose. Thats just a stopper as the hose/calliper is prefilled.
Just cut at the correct point, fit new barb and olive and fir to the lever.
