I have a new (to me) wheelset with extralite Cyber Hubs. When fitting the shimano DA 12-Speed Casette, there is not one, but multiple positions ins which the Casette fits.
Furthermore, in each position there is slight play.
What am I doing wrong?

That’s the wrong driver for that casette. You need Microspline.
