Hi, I'm preparing for bikepacking trip and one of my spokes fell of today on a perfectly flat road. Should i change the wheel? Could it happen again? Is it ok if I continue to ride without a spoke?

You mean it broke.
You should replace it.
Missing spoke could lead to wheel go out of true and especially because you will be loaded with additional weight, that could lead to a few other spokes to break/snap.
Replace it, cycling on it will break more spokes. Looks like a strange break though, mine always broke at the hub. Also make sure to put the right amount of tension on it after you replace it.
