I’ve cannot take off my tire for the life of me (yes it’s deflated). There appears to be some sort of ‘glue’? The tire is the ‘Tufo S33 Pro’. Can anybody help me?

That is a tubular type tire. It’s glued or taped to the rim with a very strong adhesive
Look up a tutorial for how to remove a tubular, but essentially you’re gonna have to warm the tire up a bit, and try and pry the tire from the rim, clean up all the mess, and then prep the tire in order to glue/tape on another tire.
It’s not the easiest thing in the world, so good luck! Probably worth a visit to your LBS and pay them to do it for you
Tubular tires are nice to ride on and a lot of people do, but they’re a huge hassle when it comes to changing them out. Unfortunately, only tubular tires will work on those rims, you can not fit a normal tire and tube to it.
You should ask yourself if you want pay the price to continue using tubular tires, or if you’d rather try and sell the tubular wheels and buy a new set of normal “clincher” wheels
