Considering these wheels as an upgrade from the stock Giant SR-2 disc wheels on my 2016 Giant Defy 2.

Could I throw my 10 speed cassette and disc brake rotor on there without too much of an issue? I’d just need a 1.85mm spacer for the cassette?

Inner rim width goes up quite a lot, from I believe 17 mm to 22 mm. So you can better fit larger *tires and they should be marginally more aero. And look better. Don’t know what the clearance on a 2016 defy might be though.
Apart from that I don’t think it’s much of an upgrade.
Tiagra 10s should work, exactly as you say. Rotors too, they both should be center lock right?

I’ve upgraded wheels a few times, and a few hundred grams on wheels really changes everything. Depend on your riding, I am riding in stop/go city traffic and 200g means a lot. A few minutes faster on 20km, but more importantly it feels much better
