Switching to tubeless with new wheels after using TPU for the past year. After seating the tire without sealant/tube, noticed the leak from this puncture. I got the Orange endurence seal to inject from the fillmore valve. So the ultimate question is would this seal or not?

Those tires are dry rot and cracked all over. It may seal for now but there’s not a lot of life left in tire. I’d replace it personally.
Yes, it will seal, it the size doesn’t increase.
But that tire is doomed, you can see lots of cracks close to the base of the thread.
