
So I’ve changed multiple wheels and flat tires over the years, but I have no clue how to get this wheel off my cervelo!! Derailleur pulled all the way back plus deflated the tire to give it more wiggle room… any help would be appreciated before I take it into the bike shop!

A dumb question, but tricky to tell from that photo: have you actually removed the thru axle? Just a thump on top of the wheel if it’s binding once that’s out will usually do the trick.
You need to remove the axle entirely from the hub.
Keep unscrewing it until you can slide it out, then give it a bump with your hand downward.
Your RD is mounted incorrectly, the B-link should be approximately horizontal.
