Had a major hole in the tire and put a big tubeless Plug into it. There is no air or milk escaping but im unsure if i should replace the tire. Its a GP 5000 AS TR in 35mm

I’ve only ever ridden with tubed buttplugs. Is there a noticeable difference in feel?

That big of a hole on that narrow of a tire I’d definitely replace.

Patch it from the inside and keep riding.

I’d ride it and carry a tube with me just in case. Those are costly tires to replace.

How large was the hole?
Unless it was significantly larger than ~5mm, I would patch it and never think about it again. If it were smaller than ~5mm I’d probably leave the plug in and never think about it again unless the plug was so long inside the tire that I could feel the balance being off.
