I’m assembling my new Endurace and am a little confused about the width adjustment of the cockpit.
The manual says to put 2 screws, but on the widest setting (leaving 2 holes for max gap as per the manual) there is nothing for the inner screws to grab on, only the outer ones hold the handle in place. The “no gap” is even more confusing, as it suggests to not adjust the width at all but put the handle all the way in.
Am I missing something or is one screw enough?

You are over extending. The widest setting is the one that still allows for both bolts.
That manual is seriously weird. More questions than answers in those two images you circled. My 2025 Aeroad came with clearer, more common sense instructions.
As for one or two screws holding the drops, I think the biggest issue with one screw per side (beside the point it’s not meant to be mounted that way), would be that the overlap between cockpit and bar end would be too small and could lead to failure. If my CP0048 cockpit is anything to go by, the tolerances between the sliding parts are quite exact and tight enough, that one screw could probably hold it in place, but the additional leverage would be unsafe for the smaller insertion depth.
The bar tape is wrapped around the outer holes for the narrow option normally. So you should have three holes on the drops free and on top of the drops you should also see width indicators. You need two screws.
