Currently in Quito, Ecuador and noticed that on one of my tyres the rubber has come away from the layer below. It holds air fine.
I’m having a lot of trouble finding a replacement. Nobody seems to have 650B/27.5” tyres here in a width that will fit my bike.
Is this going to be possible to fix? Could I get a shoe repair place to glue it?

That’s a big No in the US
In Central America I would definitely find a crafty person to glue that up and roll with it. What else ya gonna do? Grab some super glue to take with you after the repair.
Glue and sew it. On expedition bikepacking trips I carry a sew kit with me.
Good advice above. Go to an automotive or tire place and see if they can boot it. 2nd best bet: go to a cobbler.
If not: vulcanizing Glue (like from your patch kit) or rubber cement, plus sewing.
I’ve had remarkably good luck with strong tape around the tire & rim in the past, but only for an emergency.
Front or rear? If it were to blow out after your repair I’d rather it be installed as the rear tire instead of the front.
Sew it with fishing line. Strongest patch you can find on the inside then and fashion a boot of some sort to reinforce it further. Keep your pressure low then and I’d say it’ll see you til you get a replacement
