I was building a spectral 5, but I think I did not put the cable in the dropper seat correctly.
I went to remove the screw from the dropper remote, but it will not release. I can tighten and loosen it, but the screw will not come undone.
Any ideas how to remove it?

Are you talking about the pinch bolt that holds the cable, or the one that goes into that horizontal slot?
The pinch bolt is self explanatory and probably the one you want to remove honestly. If you need to remove the full lever then those Shimano lever screws have 2 sets of threads.
The initial threads tighten to connect the lever to the ISPEC-EV standoff. The second set of threads holds it securely in position in the horizontal slot.
Once you back the first threads all the way out, pull outward in the remote while you continue to unscrew it which will engage the second set of threads and allow you to remove it from the mounting point entirely.
You can see a pic of it here, notice the smooth part between the two sections of threads — my guess is you’re just between the two threads and the bolt is just spinning freely: https://pitcrewcycles.com/products/shimano-xtr-sl-m9000-i-spec-ii-rapidfire-plus-lever-integration-bolt-unit-m5-x-12-y01u98060-y01u98060
