Bike won’t shift into biggest and second biggest cog. Much to my surprise, the adjustment screw touches and does nothing. What am I looking at here?!

Shift into the biggest gear and adjust the screw that is stopping the deraiuler from moving. It is most likely set incorectly and the screw prevents it from going up.
Has the cable slipped? Does it go totally slack on the small cogs? Will it go onto the big cogs if you push it with your finger?
On the smallest cog, your cable is likely slack and you are losing range on the derailleur.
Here’s how I adjust derailleurs:
1. Unfasten the cable completely. Adjust the small cog stop screw so the chain runs smoothly on the smallest cog.
2. Ensure the shifter is in the small cog position and the cable tension adjuster on the derailleur is screwed in almost all the way (I use roughly one turn from fully screwed in).
3. While holding tension on the cable (with pliers), clamp the cable on the derailleur body with the clamping screw or nut.
4. Use the cable tension adjuster on the back of the derailleur to increase tension on the cable until the derailleur just starts to move.
5. Shift up the cassette into the largest cog (carefully). The large cog stop screw should be close to a stop at this point. Adjust the large cog stop screw until the derailleur just starts to move. If the derailleur does not go into the largest cog, then unscrew the stop screw until it can.
6. Shift through all the gears. Should be fairly well adjusted at this point, but you might need to play with the cable tension (with the cable tension adjuster on the derailleur) to get perfect gear indexing.
